Generation of protocols, data acquisition and analysis: Specific voltage clamp protocols are detailed in the relevant Results sections. Voltage clamp command signals were generated by a Cambridge Electronic Design (Cambridge, United Kingdom) 1401 interface and WCP (a program written and supplied free of charge by John Dempster of Strathclyde University). Data were recorded directly online with a Viglen EX personal computer. Membrane currents were sampled at 2 kHz. ICa,L amplitudes were measured as the difference between the peak inward current at the start of the test pulse and the steady state value at the end of the pulse. Graphs were drawn using FigP for Windows software (Biosoft, Cambridge, United Kingdom), and statistical analysis was performed with Systat for Windows (Systat Inc, Evanston, Illinois, USA). Each concentration of drug was applied to AVN myocytes from at least two hearts, and each experimental observation was repeated on at least four cells (unless otherwise indicated). Mathematical fits of data were performed with FigP for Windows. Dose-response data were fitted by the following Langmuir function:
