A new antiarrhythmic principle (part 4). ANIMALS AND METHODS

Intracellular electrophysiology: In additional experiments, papillary muscles from the right ventricle were obtained from male guinea pigs weighing 250 g, killed by a sharp blow to the neck and exsanguinated; hearts were excised within 1 min and transferred to a 10 mL organ bath with gassed Tyrode’s solution. Papillary muscles were carefully prepared and paced at 1 Hz via platinum electrodes, bathed in the same Tyrode’s solution as described above, at 31 °C, 10 mL/min and subjected to 10-6 mol/L antiarrhythmic peptide AAP10.Action potentials were recorded using glass capillaries (made from type GC 150 F10 capillaries, Clark Electromedical Instruments, Pangbourne, UK; electrode resistance 20 MQ, tip diameter 1 |j,m, pipette puller Mikroelektrodenziehgerat DC, Hugo Sachs Elektronik, Hugstetten, Germany) filled with 3 mol/L KCl using standard electrophysiological equipment (1.5 mL Steiert organ bath; HSE-Mikroelektrodenver-starker, Hugo Sachs Elektronik) with a computer-assisted data acquisition and analysis system (TIDA, FMI GmbH, Egelsbach, Germany). This system has been described in more detail previously . The muscles were stimulated at 1 Hz with 1 ms rectangular pulses of double threshold (Stimulator T, Hugo Sachs Elektronik) and equilibrated for 30 mins with Tyrode’s solution (superfused at 10 mL/min). After equilibration 1.0 |J.mol/L AAP10 was added to the superfusion solution for 30 mins, followed by a 30 min washout period.
